Where do I start with databases?

Hi Folks,

I use VS2010 and would like to create and manage a very large database. I have used MS Access in the past but I need to store more than 2GB of data so I guess it is time to upgrade. I really only need a desktop database that will not be accessed externally. Can anybody suggest where I should start? My new database may be upwards of 5GB in size.
